De-Dup Files On Multiple Hard Drives?

I have 6 internal hard drives in a PC. Four (4) of those drives contain music folders (among other folders). The music files total approximately 8TB.

I would like to De-Dup 6 different folders simultaneously that are contained on these 4 drives. Can I configure that to work? If so, because of the 8TB size, would it be recommended to do?

You can add multiple locations to PerfectTUNES, it might take a while to work through the collection if 8TB.
